ELI.H.A Echinococcus

Indirect hemagglutination test for serodiagnosis of Hydatidosis.

Description

Intended Use

Hydatidosis or echinococcosis is a parasitic disease caused by the larvae (hydatids) of a cestode of theEchinococcus genus. The life-cycle of Echinococcus granulosus requires both definitive and intermediate hosts. In general, the dog is the definitive host, with sheep and in rare cases humans being the intermediate host.

The hydatids are most frequently found in the liver (50 to 70%) and then the lungs (25 to 40%). E. granulosushydatid infection is characterized by a slow and insidious disease progression. Infection is relatively asymptomatic, with biological diagnosis resting primarily upon the detection of antibodies.

ELITech developed the ELI.H.A Echinococcus, an indirect hemagglutination assay, for the quantitative determination of anti-Echinococcus granulosus antibodies in the serum. It is rapid, easy-to-use and easy-to-read with results available within 2 hours and reliable overnight.

Principle

Sheep red blood cells are sensitized with Echinococcus granulosusantigen.

Diluted serum is mixed with sensitized sheep red blood cells. If anti-Echinococcus granulosus antibodies are present in the serum, sensitized red blood cells will agglutinate, resulting in a cloudy red/brown deposit coating the well. In the absence of specific antibodies, sensitized red blood cells will not agglutinate, resulting in a ring-like deposit at the bottom of the well.

Non-sensitized red blood cells are also available in the kit. They ensure the specificity of the reaction by allowing identifying any interference from the natural anti-sheep agglutinins (Forssman heteroantibodies, infectious mononucleosis antibodies…).

The reaction is carried out in a U-microplate.

Each kit allows 102 tests to be carried out or 17 reactions of 6 dilutions. Results are obtained within 2 hours and are reliable overnight.

Easy-to-Read and Easy-to-Interpret Result :

Titer Interpretation
<1:80 Non-significant reactionProbable absence of hydatidosis infectionRenew the test 2 to 3 weeks later and also carry out an electrosyneresis or an immunoelectrophoresis test
1:80≤T≤1:160 Doubtful reactionRenew the test 2 to 3 weeks later and also carry out an electrosyneresis or an immunoelectrophoresis test
≥1:320 Significant reaction in favor of progressive hydatidosis
